What is the chromatograph for?
The chromatograph first separates the mixture with a chromatographic column, and then sequentially detects the separated components with a detector. The diameter of the chromatographic column is several millimeters and it is filled with solid adsorbent or liquid solvent. The filled adsorbent or solvent is called stationary phase, and the sample is separated in the chromatographic column according to the different thermodynamic properties.
Classification of chromatograph
Mature chromatographic instruments mainly include gas chromatograph and liquid chromatograph. Gas chromatograph has the advantages of high resolution, high speed, high sensitivity and good selectivity. However, gas chromatograph can only be used to separate and detect vaporized substances.
There are many kinds of solvents available in liquid chromatography, which can change the polarity, pH value and concentration of solvents and adjust the difference of sample distribution between two phases, thus effectively improving the separation conditions and achieving the ultimate goal of improving separation.
