RFLP (Restriction Fragment Length Polymorphism), as the first generation of molecular biology markers, has been widely used in a variety of biological disciplines since its inception, but its use in plant resistance research is only in recent years.

RFLP can localize and isolate resistance genes in plants, and using RFLP technology, polymorphisms in the nuclear genome or chloroplast genome, especially the latter, can be seen directly from the electrophoretic profiles of the enzymatically cleaved DNA if pure DNA can be extracted, and the differences in the evolutionary level of resistance divergence between different levels of species in polluted environments can be determined within and between populations using this method. The RFLP technique can be used for genotyping as well as for the study of microbial diversity in different environments.

Advantages and disadvantages:

RFLP is characterized by low-copy coding sequences and is very stable, but RFLP is cumbersome and costly, and is not suitable for large-scale 'molecular breeding', and RFLP needs to be converted to PCR-based markers in plant molecular marker-assisted breeding.

Compared with nucleic acid sequence analysis, RFLP can eliminate many of the very tedious processes in sequence analysis, but relative to RAPD, RFLP method is more time-consuming, laborious, and requires a variety of enzymes for DNA cleavage, transmembrane, and the preparation of probes and other steps, and only a single copy of the genome sequence for identification. However, RFLP is superior to RAPD in that it can be used to determine whether a polymorphism is produced by the parent, and whether the mutation is caused by a base mutation or inversion, or by a deletion or insertion.
