What are the advantages of serum acetate fiber membrane electrophoresis compared with filter paper electrophoresis?

Compared with filter paper, cellulose acetate membrane electrophoresis of serum protein has the following advantages.

The adsorption of (1) cellulose acetate membrane on protein sample is very small, and there is no "tailing" phenomenon. After dyeing, the background can be completely decolorized, and various protein staining bands can be clearly separated, which improves the accuracy of determination.

(2) save time quickly. Because the hydrophilicity of cellulose acetate membrane is smaller than that of filter paper, the membrane contains less buffer and the electroosmosis effect is small, and most of the current is conducted by the sample during electrophoresis, so the separation speed is fast and the electrophoresis time is short. Generally, electrophoresis takes 45-60 minutes, and with dyeing and decoloration, the whole electrophoresis can be completed in about 90 minutes.

(3) High sensitivity and less sample consumption. Serum protein only needs 2μl serum, even if the sample size is as small as 0. 1μl, a clear separation band can be obtained for samples containing only 5μg protein. Clinical medical tests use this to detect the changes of trace abnormal proteins in pathological state.

(4) Widely used. Some protein are difficult to separate by paper electrophoresis, such as fetal gamma globulin, lysozyme, insulin, histone, etc., and can be well separated by cellulose acetate membrane electrophoresis.

(5) After electrophoretic dyeing of cellulose acetate membrane, it can be made into transparent dry plate after being soaked in mixed solution of glacial acetic acid and ethanol or other solutions, which is beneficial to scanning quantification and long-term preservation.
