What is the core of column separation system?

The core of separation system is chromatographic column, which is used to separate multi-component samples into single components.

Chromatographic columns are divided into two categories: packed columns and capillary columns (open columns).

Packed column consists of column tube and stationary phase in the column. Packed column is characterized by simple preparation and many kinds of stationary phases, which can solve various analytical problems. The main disadvantage is low column efficiency.

Capillary column can be divided into hollow capillary column, packed capillary column and porous layer capillary column. Capillary column is characterized by high column efficiency.

Column efficiency is simply the ability of chromatographic column to separate two substances in chromatography.

The higher the column efficiency, the better the separation effect and the better the peak shape. On the contrary, the separation effect and peak shape are poor, and even the two substances can not be completely separated. The column efficiency of chromatographic column is related to packing, column length and column inner diameter. Generally speaking, the finer the packing, the greater the length-diameter ratio of the column and the higher the column efficiency.
