About the detection of Salmonella
Operation method of Salmonella Fiona Fang test piece:
1. Sample treatment: Take 25mL(g) sample and put it into a sampling tank or homogenization cup filled with 225mL diluted sterile phosphate buffer (or physiological saline) to prepare a sample homogenate of 1: 10.
2. Inoculation: Put the test piece of Salmonella (BS205) on a flat experimental platform, uncover the upper membrane, suck the 1ml sample with a sterile pipette, slowly and evenly drop it on the paper, then slowly cover the upper membrane and let it stand for about 10s to solidify the culture medium, and inoculate two pieces for each sample. At the same time, a blank negative control was made.
3. Culture: Stack the test pieces together and put them back into the original self-sealing bag for sealing. The transparent surface is horizontally placed in a constant temperature incubator, and the number of stacked pieces shall not exceed 12. The culture temperature is 36℃ 65438 0℃ and the culture time is 65438±05 ~ 24h.
Interpretation of results:
Observing the test piece, it was found that the purple colony was Salmonella. The blue colonies are other flora. For samples with positive colonies, it is best to use other more reliable methods for verification. If there are no conditions, at least another sample should be taken for repeated inspection.
