Advantages and disadvantages of in vivo culture and in vitro culture when monoclonal antibodies are used.
Up to now, the method of producing monoclonal antibodies in animals is usually adopted.
Advantages: The method is simple and economical.
Disadvantages: Ascites are often mixed with various miscellaneous proteins of mice (including Ig), so in many cases it needs to be purified before it can be used, and there is also the danger of polluting animal viruses, so it is best to use SPF mice.
(2) The method of producing monoclonal antibodies by in vitro culture.
Advantages: It is convenient to concentrate and purify monoclonal antibodies.
Disadvantages: Special equipment is required. Moreover, the amount of monoclonal antibodies prepared by this method is extremely limited, which is not suitable for large-scale production of monoclonal antibodies. In order to prepare a large number of monoclonal antibodies in vitro, it is necessary to cultivate a large number of hybridoma cells (high density). The more cells per unit volume, the longer the cell survival time, the higher the concentration of monoclonal antibody and the greater the yield.
